A 15 year old male patient presented to pulmonary medicine OPD with 1 week history of cough with blood tinged expectoration. Sputum sample was sent for microbiological examination.
Staining method- auramine phenol staining of sputum smear. Staining shows bright brilliant bacilli against dark background in auramine phenol staining.
The staining method is SPUTUM SMEAR FLUORESCENCE MICROSCOPY.
It is a phenol Auramine O and KMnO4 stain.
Auramine O is a yellow fluorescent dye used to stain acid-fast bacteria in sputum or in paraffin sections of infected tissue. Potassium Permanganate dissolves in water to give deep purple solutions.
